همراهی بین پلی‌مورفیسم (765G>C-) ژن سیکلواکسیژناز 2 و سطح سرمی پروستاگلندینE2 در مبتلایان به دیابت نوع دو

زمینه و هدف: سیکلواکسیژناز 2 (COX-2) یک آنزیم پیش‌التهابی است که مهم‌ترین محصول آن پروستاگلندینE2 (PGE2) است که می‌تواند در ایجاد دیابت نوع دو نقش داشته باشد. یکی از عوامل موثر بر فعالیت یا بیان سیکلواکسیژناز 2، پلی‌مورفیسم‌های ژنتیکی است. این مطالعه به منظور ارزیابی همراهی بین پلی‌مورفیسم (-765G>C) ژن سیکلواکسیژناز 2 و سطح سرمی پروستاگلندینE2 در مبتلایان به دیابت نوع دو انجام شد.

روش بررسی: این مطالعه مورد شاهدی روی 50 بیمار مبتلا به دیابت نوع 2 و 50 فرد سالم مراجعه کننده به مرکز آموزشی درمانی شهید صیاد شیرازی گرگان انجام شد. سطوح سرمی FBS، HbA1C و PGE2 با استفاده از کیت الایزا و پلی‌مورفیسم ژن به‌روش ARMS-PCR تعیین گردید.

یافته‌ها: همراهی پلی‌مورفیسم765G>C ژن COX-2 با هر دو متغیر دیابت و PGE2 از لحاظ آماری معنی‌دار نبود. سطح پروستاگلندینE2 در افراد مبتلا به دیابت (43.42±31.2) در مقایسه با گروه سالم (17.27±6.38) بالاتر بود (P<0.05). سطح پروستاگلندینE2 در افراد با ژنوتیپ CG (41.15±30.99) نسبت به افراد دارای سایر ژنوتیپ‌ها به‌طور معنی‌داری بالاتر بود (P<0.05).

نتیجه‌گیری: پلی‌مورفیسم 765G>C آنزیم سیکلواکسیژناز 2 به‌عنوان یک واسطه التهابی بر خطر دیابت یا پیچیدگی‌های آن اثری نداشت.
